BACKGROUND/AIM: Hemorrhoidectomy is often associated with significant postoperative complications that may result in slow wound healing. The traditional Chinese medicine (TCM) compound Sophora flavescens (CSF) has shown efficacy on many inflammatory disorders. The aim of the present study was to examine the efficacy of CSF on wound healing in a rat model of perianal ulceration. MATERIALS AND METHODS: A rat model of perianal ulceration was induced by subcutaneous injection of 75% glacial acetic acid. The animals with induced perianal ulcer received topical treatment of low, medium, and high doses of CFS twice daily. Potassium permanganate (PP); 0.02%) was given to the animals for comparison. Macroscopic and histological assessments of the ulcerated area were performed after treatment. The expression of pro-inflammatory cytokines prostaglandin E2 (PGE2) and interleukin-8 (IL-8) was detected by immunohistochemical analysis. RESULTS: Topical administration of medium- and high-dose CSF significantly enhanced perianal ulcer healing as compared to the untreated control (p<0.05). The macroscopic ulceration score was significantly reduced only in the high-dose CSF-treated group as compared to the control (p<0.01). All doses of CSF and PP ameliorated histological damages in the rats with induced perianal ulceration. High-dose CSF or PP significantly reduced the expression of PGE2 and IL-8 as compared to the control (p<0.01). No treatment-related toxicity was found in either the CSF- or the PP-treated mice. CONCLUSION: CSF enhances wound healing in a rat model of perianal ulceration. The inhibitory effect of CSF on pro-inflammatory cytokines PGE2 and IL-8 may be involved in the mechanism of enhanced wound-healing.

RESUMO

Twenty-seven German shepherd dogs with perianal fistulas and histological evidence of colitis were entered in a prospective clinical study to investigate the association between perianal fistula and colitis. Additionally, the response of perianal fistula disease to immunosuppressive doses of prednisone and an alternative protein diet was evaluated. All 27 cases completed the treatment protocol, and perianal fistulas completely resolved in nine (33.3%) cases, improved in nine cases, and remained unchanged in nine cases.

Two cows with synchronised oestrous cycles were housed with a Friesian bull. During the midcycle period of the cows, one cow was given an intradermal injection of adrenaline in the perineal region. This caused local sweat gland discharge. The other cow was treated with water. The number and types of bull-to-cow behaviours were recorded before, during and after the day of treatment. This was repeated at three-weekly intervals for a total of four times. On the day of treatment the bull directed a greater proportion of olfactory behaviours towards the adrenaline-treated cow than the other cow (P less than 0.05). There was no difference between the cows before or after the day of treatment. The above experiments were repeated with a Hereford bull who showed the same response as the Friesian for the first two but not the last two experiments. The increased proportion of bull olfactory behaviours elicited by an increased perineal skin gland discharge, adds support to our hypothesis that the perineal skin glands are the source of an oestrous pheromone in the cow.

RESUMO

In vivo and in vitro experiments on the endocrine relationships of epidermal glands in the tokay Gekko gecko, and the common house gecko Hemidactylus bowringii are reported. The results show that certain aspects of beta-gland differentiation involve a synergistic action between androgens and those hormones responsible for controlling the normal shedding cycle, while other aspects are solely under androgenic control. Pre-anal organ activity appears to be solely under androgenic control.
